Die Rolle des genetischen Risiko-Scores bei der Erkennung und Prävention der Alzheimer-Krankheit

Die Alzheimer Krankheit ist eine neurodegenerative Krankheit, die 1 von10 älteren Personen im Alter von 65 Jahren und älter betrifft. Die Vererblichkeitvon Alzheimer wurde auf 74% geschätzt. Das ε4-Allel vom Apolipoprotein E ist ein starker genetischer Risikofaktor der Alzheimer Erkrankung. Jedoch sind nur 40 bis 65 Prozent der Alzheimererkrankten ε4 Träger und daher muss es mehrere assoziierte genetische Loci geben. In den letzten Jahren identifizierten große genomweite Assoziationsstudien mindestens 21 zusätzliche genetische Loci, die mit Alzheimer assoziiert wurden, was die Entwicklung des genetischen Risikoscores (GRSs) erhöhte. GRSs rechnen das genetische Risiko unter  Berücksichtigung vieler kleiner Wirkungen verschiedener genetischer Loci. Alzheimer GRSs nutzen Alzheimer assoziierte einzelne Nukleotidpolymorphismen (SNPs) und korrespondierende Assoziationen, um den Risikoscore GRS zu bilden.  Durch die frühe Identifizierung von Personen mit Alzheimerrisiko vor der Manifestation der Erkrankung, haben GRSs das Potential,  zu einer  individualisierten und genauen Alzheimerrisikoprognose beizutragen. Im Rahmen der Dissertation wird ein Alzheimer GRS entwickelt und validiert um die gemeinsame genetische Architektur zwischen der Alzheimer Erkrankung und assoziierten Krankheiten zu erforschen. Das Projekt basiert auf den Daten einer großen prospektiven Kohortenstudie: “ESTHER” (http://esther.dkfz.org/esther/).
